Oxygen metabolism analysis of a vascularized spheroid using a scanning electrochemical microscopy

We investigated the effect of vascularization on oxygen metabolism in a fibroblast spheroid. By the scanning of a microelectrode, the oxygen reduction current was monitored, which allowed us to analyze the oxygen metabolism quantitatively. The oxygen metabolism of the spheroids were determined by the spheroid radius (volume) and did not depend on whether the spheroid was vascularized or not, implying the identical activity of oxygen metabolism of the fibroblast and endothelial cells. Our system is useful for the metabolic evaluation of a three-dimensional culture model.
