Investigation into mechanism of protein resistance based on hydration states in polymer chains

This research object is an understanding of hydration states in polymer chains for inducing the protein resistance. We analyzed hydration states of 2-methacryloyloxyethyl phosphorylcholine polymer (poly(MPC)) with the same structure as phospholipid headgroups in cell membranes. The poly(MPC) hydrogels were prepared to have EWC ranging from 86 to 92 wt % and hydration states in the hydrogels are measured by DSC. Results are compared with those in the hydrogels composed of non-biomimetic reference methoxy oligo(ethylene glycol) methacrylate polymer (poly(MeOEGMA-n) (n = 4 and 8)). For the hydrogels with similar EWC, the poly(MPC) hydrogels exhibit 1.2-2.1 times higher fractions of nonfreezing water than the poly(MeOEGMA-n) hydrogels. Also, the number of nonfreezing water per the phosphorylcholine group and the ethylene glycol repeat unit is estimated as 20-26 and 2.8-4.1, respectively.
