It has been well known that peptide YY(PYY) or neurotensin (NT) has a suppressive effect on upper gastrointestinal motility as one of the important candidates for 'ileal brake'. However, physiological action of the glicentin in 'ileal brake' which is secreted from distal gut has not been previously investigated. This study was designed to evaluate the effect of the glicentin on the upper gastrointestinal motility to compare with PYY or NT. Three mongrel dogs were provided with four force transducers were placed on the gastric body, gastric antrum, middle portion of duodenum and proximal jejunum to measure the gastrointestinal motility. During 24 hours continuous recording of gastrointestinal motor activities, intravenous infusion with saline (C-group), PYY (400 pmol/kg/hr:P-group), NT (400 pmol/kg/hr:N-group) or glicentin (400 pmol/kg/hr:G-group) was started simultaneously with a standard ingestion and continuied for one hour. Significant suppressions of antral motor indexes were observed in P-group (0.683), N-group (0.423) and G-group (0.675) respectively compared to C-group (1.670). However, these hormones did not significantly alter the meter indexes of duodenum and jejunum. The durations of postprandial phase did not differ significantly among all groups. These results suggest that the glicentin has a suppressive effect on postprandial gastric motility similar to the effect of PYY or NT.
