Design of enzymatic-responsive artificial molecular chaperone

We synthesized alkyl group bearing maltopentaose as an amphiphpilic primers of enzymatic polymerization of amylose. For example, dodecyl group primer (DA-primer) was formed micelle in aqueous solution. The enzymatic polymerization by phosphorylase b of DA-primer proceeded 4 times faster than that of maltopentaose. The association and dissociation of DA-primer was controlled by enzymatic reaction. Chaperone like activity was examined by using DA-primer micelle polymerization system. DA-primer micelle trapped refolding intermediate of denatured CAB. Denatured CAB in micelle was refolded upon dissociation of the micelle with proceeding the enzymatic polymerization of DA primer. The enzymatic activity was reached over 50 % by 120 min polymerization. We could construct novel artificial molecular chaperone system by enzymatic response of DA-primer micelle.
