Construction of isogenic cell lines expressing human and rat angiotensin II AT1 receptors by cre-mediated site-specific recombination

The Cre site-specific recombinase of coliphage P1 is functional in eukaryotic cells and can precisely direct integration of exogenous DNA to a predesignated chromosomal target containing the Cre recognition site termed lox. To exploit this system for the production of isogenic cell lines expressing heterologous genes, we have constructed cDNA lox targeting vectors. Using a CHO cell line containing a solitary lox target, we have inserted into the genome a single copy of the angiotensin AT1 receptor gene from both rat (two isoforms) and human (one isoform). Isogenic, single-copy cell lines that produce a functional angiotensin receptor and that differ only in the structural AT1 gene being expressed can thus be generated.
