Composition of free form glycopeptides isolated from bovine colostrum

Free peptides were isolated from bovine colostrum taken immediately after parturition by combining 12% (w/v) tricholoracetic acid (TCA) extraction and reverse-phase (RP) preparative chromatography with silica gel linking octadesyl residues. The average quantity of free peptides in bovine colostrum was ca. 350 mg/l (three trials). Two major glycopeptides. A and B, were isolated from the free peptide fraction by gel filtration chromatography with a Sephadex G-25, followed by preparative HPLC. Molecular weight of the glycopeptides A and B was estimated by HPLC to be 7·9 kD and 8·3 kD, respectively. Amino acid and sugar composition, and specific affinity for lectins were also determined for the glycopeptides. A complex type of sugar chain: NANA-Galβ1-4{(GlcNAcβ1-2-Mana1-6)(GlcNAcβ1-2Manα1-3)} (GlcNAcβ1-4)-Manβ1-4GlcNAcβ1-4GlcNAc → Asn and NANA-Galβ1-4GlcNAcβ1-2{(Manα1-6)(Manα1-3)}-Manβ1-4GlcNAcβ1-4(Fucα1-6) GlcNAc → Asn was estimated to bind N-glycosidically to the glycopeptides A and B, respectively.
