    To study mammalian enzymes for repair of oxidatively dam a cod DNA. < DNAs encoding homologues (inNthl and hNTHl) of Kxchrrtrkiti i-nli cndonuc-Icasc III (endo III; nth gene product) were isolated from moue arid human cDNA libraries, respectively, by screening with mouse cDN'A fragments am pli fied based on an information reported in KST Data Hases. inNthl and h.\ IHl cDNAs were 1025 and 1028 nucleotides long, and encode the cn/yme proteins consisting of iiOO and'304 ainino acid residues, respectively. Deduced amino acid sequences of both proteins exhibit significant homologios to thohf of cudo III and their oukaryotic homologues, including two motifs contesting, of a iielix hairpin-helix arid an iron-sulphur [4Fe-4S] cluster. niNthl niKNA (OT hNTHl niKNA) of approximately 1 kb was expressed in all mouse (or human) tissues analyzed. m.Nthl gene was cloned, and .shown to consist of (i exous spanning 6.09 kilobases. The boundaries between exon and intron follow the t.i'1 /A(! i u le.
