Analysis of the human metallothion-II gene in age-related macular degeneration

Purpose. Several authors have reported a decrease of metallothionein (MT) concentration in macular RPE due to aging and it may concern to age- related macular degeneration (ARMD). The purpose is to examine the expression and sequence of the MT gene in the fibrovascular membrane in patients with ARMD and compare it to those of proliferative membrane in pauents with proliferative vitreoretinopathy (PVR) or normal RPE. Methods. mRNAs from proliferative tissues of ARMD , PVR and from normal RPE were extracted and reverse tanscriptase-polymerase chain reaction (RT-PCR) was performed. A part of these samples were subcloned and used for sequence analysis. Results. No different amount of MT- II gene expression was observed in these samples. However we found four different polymorphism in MTn gene. Also several angiogenetic factors such as bFGF gene was expressed abundantly in the ARMD samples compared to other ones. Conclusions. We found no different amount of MT- II gene expression between ARMD and PVR or normal RPE, but we found four different polymorphism in the MT- II gene. Though several isoformes of MTs have been reported, further examination may be necessary to elucidate the participation of other MTs for ARMD as well as the meaning of the polymorphism in the MT- II gene.
