Single molecule measurement of polymer dynamics by atomic force microscopy

    Abstract

    The technique of force measurement by atomic force microscopy (AFM) have enabled us to investigate mechanical and structural properties of single macromolecules directly. In this study, we constructed an external piezo-scanner control system and a data acquisition system for AFM, and thereby realized dynamic measurement of mechanical properties of a signle polymer chian. The response of the single polymer chain to externally applied oscillation reflected the viscoelasticity of it. This technique can lead to the detailed evaluation of intra-molecular dissipation process of a single molecules.
