Liver targeting of plasmid DNA with a cationized pullulan for tumor suppression

Abstract

The preparation of a non-viral gene carrier to specifically enhance the gene expression of plasmid DNA of NK4 in the liver was discussed. An experimental mouse model of liver metastasis was prepared by intravenous injection of RLmale 1 lymphoma cells. The mouse survival and serum glutamic pyruvic transaminase (GPT) activity were examined at different time intervals. It was observed that when the in vitro gene expression was examined in the presence of asialofetuin, the expression level was suppressed for all the complexes. It was also observed that the survival of mouse bearing RL male 1 cells was prolonged only by injection of NK4 plasmid DNA complex with spermine-pullulan.
