Function of in situ synthesized membrane protein in the presence of cell-size liposome

Abstract

Membrane proteins perform significant functions such as metabolisms or signal transductions. However, because of their hydrophobicity, it is difficult and time-consuming to isolate and purify them with keeping function. A convenient method enabling to construct pure membrane protein is awaited. We investigated in vitro membrane protein synthesis in the presence of liposomes. Connexin is a component of gap-junctions that mediate intercellular communication. Cell-free membrane protein synthesis for connexin43(C×43) was carried out and functional protein(C×43) containing liposomes was directly obtained. The bidirectional transfer of water-soluble dye, calcein through gap junctions was observed between C×43-expressing liposomes and cultured cells. The efficiency was estimated about 30 %. These results also show the C×43-liposome's possibility for novel cytosolic delivery system.
