N2 - This paper reports a high-throughput and rapid method for a fluorescence polarization immunoassay (FPIA) of H5-avian influenza virus (H5-AIV) and anti-H5-AIV antibody using a portable FP analyzer with a microdevice. For both assays, a fluorescein-labeled recombinant fragment of H5-hemagglutinin was prepared. Although the labeled fragment had low molecular-weight, it served as a tracer for the FPIA. Also, a microdevice having multiplex microchannels was designed, allowing simultaneous multiplex analysis at a small sample volume. Consequently, the virus and the antibody in several samples were successfully detected rapidly in the same format.
