Con a receptive sites on the surface of photoreceptor outer segments of lampetra japonica: TEM quantification by thin-sectioning and freeze-etching

Abstract

Lamprey photoreceptor outer segments were labeled with ferritin-Con A by pre-embedding labeling. Based on the ferritin particles associated with the membrane on the TEM of thin sections, Con A receptive sites were calculated as 487/μm2 in the long-cell's vs. 1,145/μm2 in the short-cell's outer segment. By counting on the freeze-etching replicas, the density came out as 331/μm2 vs. 1,113/μm2 correspondingly. Counting on the TEM micrograph will be useful for quantification of the affinity reactions on the plasma membrane.
