A novel lipid hydroperoxide-derived modification to arginine-containing peptides

Abstract

A method was proposed to provide dosimeters of exposure to lipid hydroxyperoxide modification to arginine containing peptides. The 4-Oxo-2-nonenal was reacted with amino acids, peptides and proteins in transition metal ion free 100 mM MOPS buffer. The protein samples were reacted with dithiothreitol and iodoacetoamide for cisteine modification and then catalyzed by trypsin or V8 protease before LC/MS. Results show that no modification occurred on the N-terminus as b 2-NH 3 and b 3-NH 3 were observed.
