A correlation between pH and fluorescence lifetime of 2′,7′- bis(2-Carboxyethyl)-5(6)-carboxyfluorescein (BCECF) in vivo and in vitro

Abstract

The pH dependence of the fluorescence lifetime of BCECF both in solution and in intact cells of Halobacteriun salinarum has been measured with time-resolved fluorescence spectroscopy. A correlation between pH and fluorescence lifetime in vivo is different from that in vitro. The difference is ascribed to the local field produced by some protein membrane in a biological system.
