[25] Nonaprenylpyrophosphate Synthetase from Micrococcus Luteus

Abstract

This chapter describes the isolation of enzyme, the nonaprenylpyrophosphate synthetase from Micrococcus luteus. A nonaprenylpyrophosphate synthetase fraction obtained from Micrococcus luteus catalyzes the condensation of isopentenyl pyrophosphate with geranyl pyrophosphate to yield all-trans-prenyl pyrophosphates ranging in carbon number up to C45. Principle and procedure of the assay method are discussed in the chapter. The enzyme activity is assayed as usual for polyprenylpyrophosphate synthetase by determining the amount of incorporation of [14C]isopentenyl pyrophosphate into acid-labile allylic pyrophosphate.
